Viruses and mammalian chromosomes. VII. The persistence of a chromosomal instability in regenerating, transplanted, and cultured neoplasms induced by human Adenovirus type 12 in Syrian hamsters.

چکیده
The incidence of cells with karyotypic abnormalities, such as pseudodiploidy and aneuploidy, and of cells with chromosome aberrations, including breaks, fragmentations, and coiling anomalies, was estimated in regenerating, transplanted, and cultured neoplasms induced in Syrian hamsters by human Adenovirus type 12. The various neoplasms studied consisted of karyotypically heterogeneous cell populations. Numerous clones with particular chromosome complements were present, but stemlines were not a dominant feature of the neoplastic cell populations examined. A relatively high incidence of chromo some aberration persisted in all neoplasms. A possible relation ship between retention of the viral genome or part of it in the neoplastic cells and the persistence of a chromosome instability and the significance of chromosome aberration in Adenovirus12-induced oncogenesis are discussed.

Objective(s) The main objective of this study was to investigate the status of chromosome stability in 3 human-mouse hybridoma cell lines over a period of time in various passages. Materials and Methods Metaphase spreads from 3 human-mouse cell lines (HF2X653, SPMO-4 and F3B6) that had been cultured in 4 successive passages, from 1 to 4 weeks, were prepared and analyzed. Metaphase chromosome...
متن کاملIntracranial transplantation of the tumor induced by adenovirus type 12 in Syrian hamster--an experimental brain tumor model.
A method of intracranial transplantation of the tumor induced by adenovirus type 12 in syrian hamster has been described. The incidence of intracranial tumor development was 86 (90.5 %) out of 95 animals and the average survival time and tumor size at death were 15.1 days and 4.1 mm in diameter respectively. The consistency of the days of death after intracranial transplantation of the tumor wa...
متن کاملI-37: Genome Instability and DNA Damage in Male Somatic and Germ Cells Expressed as Chromosomal Microdeletion and Aneuploidy Is A Major Cause of Male Infertility
Background: Sperm chromatin insufficiencies leading to low sperm count and quality, infertility and transmission of chromosomal microdeletion and aneuploidies to next generations can be due to exposure to environmental pollutions, chemicals and natural or manmade ionizing radiation. In this project which has continued for more than 10 years and is unique in many technical aspects in Iran and in...
متن کاملDesigning E1 Deleted Adenoviral Vector by Homologous Recombination
Adenoviruses are used extensively to deliver genes into mammalian cells, particularly where there is a requirement for high-level expression of transgene products in cultured cells, or for use as recombinant viral vaccines or in gene therapy. In spite of their usefulness, the construction of adenoviral vectors (AdV) is a cumbersome and lengthy process that is not readily amenable to the generat...

Background and Objective: Discovery of genetic changes which contribute to cellular neoplastic and malignant tumor transformation is one of the major aims in oncology researches. The aim of this study was to investigate the DMBA-induced breast cancer in SD rat strains using bioinformatical methods and also to find their homologous regions in human chromosomes. Materials and Methods: In this res...
